Obtaining a stable cell line from a sample of UPS obtained during surgery to model carcinogenesis processes <italic>in vitro</italic> and <italic>in vivo</italic>.</p> <p><bold>Materials and methods</bold>. Cells were isolated from a tumor sample from a patient diagnosed with UPS. After passage 40 and stable growth, the proliferation rate, cell morphology, and growth ability in 3D culture conditions were assessed. To assess tumorigenicity, the cell suspension was injected into BALB / c nu / nu athymic mice. On the 15<sup>th </sup>day of the experiment, the tumor was removed, its volume was measured, and it was examined histologically. Using short tandem repeat (STR) analysis, the cells of the resulting line were examined for the presence of human cell markers and their uniqueness. The chemosensitivity of the cells was assessed using the resazurin test.</p> <p><bold>Results</bold>.<bold> </bold>Stable growth and formation of spheroids under 3D culturing conditions were demonstrated for the cell culture. All mice inoculated with the UPS134 cell suspension showed tumor growth, and by day 14 of the experiment its average size was 28,7 mm<sup>3</sup>. Histologically, the tumor is similar in morphology to UPS, except for a smaller amount of myxoid and less polymorphism. The cells of the obtained line were shown to contain all analyzed genetic markers of human cells, and also that the STR data of the obtained line did not coincide with the STR of other lines. For doxorubicin, gemcitabine and docetaxel, IC<sub>50</sub> (inhibitory concentration) were determined to be 15, 6.5 and 10 % of the plasma peak concentration, respectively.</p> <p><bold>Conclusions</bold>. We obtained a new cell line UPS134, which can be further used to study the pathogenesis of UPS, as well as in preclinical studies of new antitumor agents <italic>in vitro</italic> and<italic> in vivo</italic>.</p></abstract><trans-abstract xml:lang="ru"><p><bold>Введение</bold>.<bold> </bold>Ввиду ограниченного числа клеточных линий, а также высокой гетерогенности недифференцированных плеоморфных сарком (НПС) создание новых стабильных клеточных линий является актуальной задачей.</p> <p><bold>Цель исследования</bold> – получение стабильной клеточной линии из послеоперационного образца НПС для моделирования процессов канцерогенеза <italic>in vitro</italic> и <italic>in vivo</italic>.</p> <p><bold>Материалы и методы</bold>. Клетки выделяли из образца опухоли от пациента с диагнозом НПС. По достижении 40-го пассажа и стабильного роста оценивали скорость пролиферации, морфологию клеток и способность к росту в условиях 3D-культивирования. Для оценки туморогенности клеточную суспензию вводили бестимусным мышам <italic>BALB / c</italic> nu / nu. На 15-й день образовавшуюся опухоль извлекали, измеряли объем и гистологически исследовали. С помощью анализа коротких тандемных повторов (STR) исследовали клетки полученной линии на наличие маркеров клеток человека и их уникальность. Химиочувствительность клеток оценивали с помощью резазуринового теста.</p> <p><bold>Результаты</bold>. Клеточная культура продемонстрировала стабильный рост и формирование сфероидов в условиях 3D-культивирования. У всех мышей, перевитых суспензией клеток UPS134, наблюдался рост опухоли, и к 14-му дню эксперимента ее средний размер составил 28,7 мм<sup>3</sup>. Гистологически опухоль схожа с морфологией НПС, за исключением меньшего количества миксоида и меньшего полиморфизма. Показано наличие в клетках полученной линии всех анализируемых генетических маркеров клеток человека, а также то, что данные STR полученной линии не совпадали с STR других линий. Для препаратов доксорубицина, гемцитабина и доцетаксела определены показатели IC<sub>50</sub> (inhibitory concentration), которые составили 15, 6,5 и 10 % концентрации плазматического пика соответственно.</p> <p><bold>Заключение</bold>.
