To ensure the safety and effectiveness of biological drugs, it is critically important to evaluate their immunogenicity. The most commonly used enzyme-linked immunosorbent assay (ELISA) has a number of limitations, so it is important to understand the possibilities of alternative approaches, for example, the biolayer interferometry (BSI) method.</p> <p><bold>Aim</bold>. Development, validation and comparison of two methods for screening assay in the context of assessing the immunogenicity of biological drugs based on monoclonal antibodies using the example of pembrolizumab with multiple intravenous administration to <italic>Macaca fascicularis</italic>.</p> <p><bold>Materials and methods</bold>. Immunogenicity was evaluated as part of a comparative study of toxicity and local tolerance of two drugs with International Nonproprietary Name pembrolizumab following repeated administration. <italic>Macaca fascicularis </italic>(3 groups of 3 males and 3 females in each) on days 1, 8, 15 and 22 of the experiment received intravenously a control substance (buffer solution), test drug (RPH-075, JSC R-Pharm, Russia) and reference drug (Keytruda®, MSD International GmbH, Ireland) at a dose 30 mg/kg. Serum samples, were collected pre-dose and on study days 15, 22, 29, and were tested for assessment of binding antibodies to pembrolizumab using two methods, bridging ELISA and BSI.</p> <p><bold>Results</bold>. During the development and testing of methods for binding antibodies (BAb) to pembrolizumab quantification in monkey’s blood serum of by the bridge ELISA and BSI assays, it was revealed that the bridge ELISA has greater sensitivity (12.3 and 419.7 ng/mL, respectively). Determination of BAb level to pembrolizumab by ELISA revealed immunogenicity in 4 out of 12 animals; when analyzing samples by BSI, an increasing of BAb concentration was detected in only one animal.</p> <p><bold>Conclusion</bold>. Bridging ELISA is more sensitive than BSI making it the preferred method for assessing of drugs with unknown or suspected low immunogenicity.</p></abstract><trans-abstract xml:lang="ru"><p><bold>Введение</bold>. Для обеспечения безопасности и эффективности биологических препаратов критически важно оценить их иммуногенность. Наиболее часто применяемый для этого иммуноферментный анализ (ИФА) имеет ряд ограничений, поэтому значимо понимание возможностей альтернативных подходов, например метода биослойной интерферометрии (БСИ).</p> <p><bold>Цель исследования</bold> – разработка, валидация и сравнение двух методов для скринингового анализа в рамках оценки иммуногенности биологических препаратов на основе моноклональных антител на примере препаратов пембролизумаба при многократном внутривенном введении яванским макакам.</p> <p><bold>Материалы и методы</bold>. Оценку иммуногенности проводили в рамках сравнительного изучения токсичности и местной переносимости двух препаратов с пембролизумабом при многократном введении. Яванским макакам (3 группы по 3 самца и 3 самки) на 1, 8, 15 и 22-й дни эксперимента внутривенно струйно вводили контрольное вещество (буферный раствор), тестируемый препарат (RPH-075, АО «Р-Фарм», Россия) и препарат сравнения (Китруда®, МСД Интернэшнл ГмбХ, Ирландия) в дозе 30 мг / кг. В образцах сыворотки крови, отобранной до введения, на 15, 22 и 29-й дни исследования определяли содержание связывающих антител к пембролизумабу двумя методиками – мостиковым ИФА и БСИ.</p> <p><bold>Результаты</bold>. При разработке и апробации методик количественной оценки содержания связывающих антител к пембролизумабу в сыворотке крови макак методом мостикового ИФА и БСИ выявлено, что большей чувствительностью обладает метод мостикового ИФА (12,3 и 419,7 нг / мл соответственно). Определение уровня связывающих антител к пембролизумабу методом ИФА позволило выявить иммуногенность у 4 из 12 животных; при анализе проб методом БСИ увеличение количества связывающих антител обнаружено только у 1 животного.</p> <p><bold>Заключение</bold>.
