One of the virulence factors of <italic>S. pneumoniae</italic> is pneumolysin (Ply), a cholesterol-dependent hemolytic toxin that interacts with cholesterol in eukaryotic cell membranes, forms pores and leads to cell destruction and death.</p><p><bold>Aim.</bold> Immunochemical characteristics of pneumolysin and evaluation of its cytotoxic effect in the culture of Chinese hamster ovarian cells CHO-K1.</p><p><bold>Materials and methods.</bold> To obtain Ply, the <italic>S. pneumoniae</italic> serotype 3 strain was cultivated in brain heart broth, bacterial cells were pelleted by centrifugation and subjected to ultrasonic disintegration. The presence of pneumolysin in the resulting preparation was confirmed by immunoblotting with monoclonal antibodies to recombinant pneumolysin. The cytopathogenic effect of Ply was studied in a culture of proline-dependent epithelial-like Chinese hamster ovary cells CHO-K1 in RPMI-1640 medium. The hemolytic activity of Ply was assessed in a reaction with mouse erythrocytes.</p><p><bold>Results.</bold> Using ultrasonic disintegration of cells of the <italic>S. pneumoniae</italic> serotype 3 strain followed by precipitation with ammonium sulfate, a protein that formed a band on electrophoresis at a level corresponding to the molecular weight of Ply (53 kDa) was obtained. The protein possessed the ability to lyse mouse erythrocytes. The authenticity of Ply was confirmed by immunoblotting with monoclonal antibodies to recombinant pneumolysin. Ply had a cytopathogenic effect on Chinese hamster ovary CHO-K1 cells. The minimum dose of the protein that induced a toxic effect on cells, manifested in the appearance of round and small cells along with normal cells, was 16.4 μg/ml per protein. When the concentration was increased up to 65.6 μg/ml, only small round cells were detected. Further increase in concentration resulted in complete destruction of CHO-K1 cells.</p><p><bold>Conclusion.</bold> Pneumolysin can be used to develop new drugs for the treatment of pneumococcal infections.</p></abstract><trans-abstract xml:lang="ru"><p><bold>Введение.</bold> <italic>Streptococcus</italic><italic> pneumoniae</italic><italic> </italic>(<italic>S</italic><italic>. pneumoniae</italic>, пневмококк) – условно-патогенные бактерии, вызывающие воспалительные заболевания у человека. Одним из факторов вирулентности <italic>S</italic><italic>. pneumoniae</italic> является пневмолизин (Ply) – холестеролзависимый гемолитический токсин, который при взаимодействии с холестерином клеточных мембран эукариотов формирует поры и приводит к разрушению и гибели клеток.</p><p><bold>Цель исследования</bold> – иммунохимическая характеристика Ply и оценка его цитотоксического действия в культуре клеток яичника китайского хомячка CHO-K1.</p><p><bold>Материалы и методы.</bold> Для получения Ply штамм S. pneumoniae серотипа 3 культивировали в сердечно-мозговом бульоне, осаждали бактериальные клетки центрифугированием и подвергали ультразвуковой дезинтеграции. Наличие Ply в полученном препарате подтверждали с помощью иммуноблоттинга с моноклональными антителами к рекомбинантному Ply. Цитопатогенное действие Ply исследовали на культуре пролинзависимых эпителиоподобных клеток яичника китайского хомячка CHO-К1 в среде RPMI-1640. Гемолитическую активность Ply оценивали в реакции с эритроцитами мыши.</p><p><bold>Результаты.</bold> Методом ультразвуковой дезинтеграции клеток штамма <italic>S</italic><italic>. pneumoniae</italic> серотипа 3 с последующим осаждением сульфатом аммония получен белок, образующий на электрофорезе полосу, соответствующую молекулярной массе Ply (53 кДа). Полученный белок обладал способностью лизировать эритроциты мыши. Подлинность Ply подтверждена в иммуноблоттинге с моноклональными антителами к рекомбинантному Ply. Ply оказывал цитопатогенное действие на клетки яичника китайского хомячка CHO-K1. Минимальная доза препарата, индуцирующая токсическое действие на  клетки, проявляющаяся в появлении наряду с нормальными клетками округлых и мелких клеток, составляла 16,4 мкг/мл по белку. При увеличении концентрации до 65,6 мкг/мл обнаруживали только мелкие круглые клетки.
