Beta-III tubulin (TUBB3), the structural protein of microtubules, is not expressed in normal epithelium, but is frequently present in different epithelial tumors. We suggested that the screening for TUBB3 in morphologically normal tissue outside the tumor may be used for the molecular diagnostics of the local spread of cancer, which is particularly significant in case of the tumors with a high rate of local recurrence, like esophageal cancer.</p><p><bold>Objective</bold>. The quantitative assessment of TUBB3 expression in tumor and morphologically normal esophageal tissue adjacent and remote from the tumor.</p><p><bold>Materials and methods</bold>. The expression of TUBB3 in surgical biopsy specimens of tumor and morphologically normal tissue derived from 40patients after radical surgery for esophageal cancer was measured by immunocytometry.</p><p><bold>Results</bold>. TUBB3 expression was detected in 35 out of 40 studied samples of esophageal cancer, as well as in 10 out of 13 samples of morphologically normal esophagus tissue adjacent to the tumor, and in 25 out of 40samples of esophagus tissue most distant from the tumor near the resection edge. It was shown that the expression of TUBB3 in tissue specimens increases as follows: “normal tissue → adjacent normal tissue → tumor”, which basically means the existence of a positive gradient of TUBB3 expression level and intensity towards the tumor.</p><p><bold>Conclusion</bold>. The expression of TUBB3, a protein associated with tumor growth, outside the primary tumor may indicate that the tissue appearing to be normal is already affected by the malignancy. It can be used as an additional factor in tumor staging and post-operative patient management.</p></abstract><trans-abstract xml:lang="ru"><p><bold>Введение</bold>. Белок микротрубочек бета-III тубулин (TUBB3) в норме не экспрессируется в эпителиальных клетках, но присутствует во многих опухолях эпителиального происхождения. Мы предположили, что выявление экспрессии TUBB3 в морфологически нормальной ткани за пределами первичной опухоли может использоваться для молекулярной диагностики местной распространенности опухолевого процесса, что особенно важно для локально рецидивирующих опухолей, в частности, для рака пищевода.</p><p><bold>Цель исследования</bold> — количественная оценка экспрессии TUBB3 в опухолевой и окружающей морфологически нормальной ткани пищевода, прилежащей к опухоли и отдаленной от нее.</p><p><bold>Материалы и методы</bold>. Методом проточной иммуноцитофлуориметрии исследована экспрессия белка TUBB3 в хирургических биопсийных образцах рака пищевода и морфологически нормальной ткани органа 40 оперированных больных.</p><p><bold>Результаты</bold>. Экспрессия TUBB3 выявлена в 35 из 40 исследованных образцов рака пищевода, в 10 из 13 образцов прилежащей к опухоли морфологически нормальной ткани пищевода и в 25 из 40 образцов максимально удаленной от опухоли ткани пищевода вблизи края резекции. В подавляющем большинстве случаев уровень и интенсивность экспрессии TUBB3 у каждого больного в опухоли превышали те же показатели в отдаленной морфологически нормальной ткани. Показано, что в ряду «норма → норма вблизи опухоли → опухоль» экспрессия TUBB3растет, т. е. прослеживается положительный градиент уровня и интенсивности экспрессии белка по мере приближения к опухоли.</p><p><bold>Заключение</bold>. Экспрессия ассоциированного с опухолевым ростом белка TUBB3 за пределами первичной опухоли указывает на молекулярную вовлеченность в опухолевый процесс визуально нормальной ткани органа и может использоваться в качестве дополнительного показателя при стадировании заболевания и определении тактики послеоперационного ведения пациентов.</p></trans-abstract><kwd-group xml:lang="en"><kwd>esophageal cancer, βIII-tubulin, flow cytometry</kwd></kwd-group><kwd-group xml:lang="ru"><kwd>рак пищевода</kwd><kwd>бета-III тубулин</kwd><kwd>проточная цитофлуориметрия</kwd></kwd-group><funding-group/></article-meta></front><body></body><back><ref-list><ref id="B1"><label>1.</label><mixed-citation>Katsetos C.D., Herman M.M., Mork S.J.
