Mammalian chitinase-like proteins are produced in the areas of inflammation and in tumors. Some members of chitinase-like proteins family are studied as potential biomarkers of tumors (glioma, prostate and ovary). YKL-39 also known as chitinase-3-like 2 (CHI3L2) is a secreted protein produced by chondrocytes. Its high expression is also found in synoviocytes, lung heart and macrophages.</p><p><bold>The aim</bold> of this study was the development of highly specific monoclonal antibodies against human YKL-39.</p><p><bold> Materials and methods</bold>. Using recombinant full-length human YKL-39 as immunogen using hybridoma technology we have generated monoclonal antibody 1B2G4, that specifically binds YKL-39 in ELISA.</p><p><bold>Results and conclusion</bold>. Obtained antibody was successfully tested in Western blot, immunocytochemistry, immunofluorescence and immunohistochemistry on FFPE sections. It was shown that the antibody binds the full-length YKL-39 protein and does not interact with other chitinase-like proteins. </p></abstract><trans-abstract xml:lang="ru"><p><bold>Введение.</bold> Хитиназоподобные белки у млекопитающих образуются в зоне воспаления и опухолевого роста. Отдельные представители семейства хитиназоподобных белков изучаются как потенциальные биомаркеры опухолей (глиомы, рака предстательной железы и яичников). Одним из охарактеризованных белков данного класса является YKL-39. Известный также как хитиназа-3-подобный белок 2, YKL-39 представляет собой секреторный белок хондроцитов, принадлежащий к семейству гликозил-гидролазы 18. Его самая высокая экспрессия наблюдается в хондроцитах, синовиоцитах, легких и сердце, а также в макрофагах.</p><p><bold> Цель исследования</bold> – получение антител к человеческому YKL-39, применимых в широком спектре методов.</p><p><bold>Материалы и методы</bold>. Используя рекомбинантный полноразмерный человеческий YKL-39 в качестве антигена, методом гибридомной технологии мы получили мышиные моноклональные антитела 1B2G4, которые специфически связываются с YKL-39 в иммуноферментном анализе.</p><p><bold>Результаты и заключение</bold> . Полученные антитела были успешно протестированы с использованием методов иммуноблоттинга, иммуноцитохимии, иммунофлуоресценции и иммуногистохимии с использованием парафиновых срезов.
